AUTONOMIC NERVOUS SYSTEM PHARMACOLOGY-CHOLINOMIMETIC DRUGS

Drug:  Nicotine 

Mechanism of action and drug effects:  Agonist at both NN and NM receptors.  Activates autonomic postganglionic neurons (both sympathetic and parasympathetic) and skeletal muscle neuromuscular end plates.  It enters CNS and activates NN receptors. 

Clinical applications:  Medical use in smoking cessation.  Nonmedical use in smoking and in insecticides. 

Pharmacokinetics, Toxicities, Interactions:  Oral gum, patch for smoking cessation.  Toxicity: Increased gastrointestinal (GI) activity, nausea, vomiting, diarrhea acutely, increased blood pressure, high doses cause seizures, long-term GI and cardiovascular risk factor.  Interactions: Additive with CNS stimulants.
